Factors Affecting Shoot Proliferation and Root Initiation of Apple ¢¥ Fuji ¢¥ (Malus domestica Borkh .) in vitro



Abstract
Factors affecting the shoot proliferation and root initiation of apple `Fuji¢¥ were described. Shoots were proliferated best on Murashige and Skoog (VIS) medium with 1.0§·/§¤ BA, 2.0§·/§¤ IBA, and 0.1§·/§¤ GA©ý. One §·/§¤ IBA was most effective for root initiation, but GA©ýwas inhibitory. Reducing the strength of MS salts was effective for root initiation, but root length was shortened. The percentage of rooting was increased with the increase of sucrose concentration up to 50g/§¤ in the continuous 16 hour photoperiod but was decreased at higher concentration than 10g/§¤ in the dark condition during auxin sensitive stage. In the continuous 16 hour photoperiod, phloroglucinol at 160§·/§¤ promoted rooting of the shoot tips while 50§·/§¤ was inhibitory, but rooting was promoted progressively with the increase of phloroglucinol concentration when the cultures were placed in the dark condition during auxin sensitive stage. Addition of activated charcoal to the medium was inhibitory for root initiation, but the root length was increased.
